In type 2 diabetes (T2DM), we hypothesized an alteration in cardiac branched-chain amino acid (BCAA) levels and the activity of branched-chain keto acid dehydrogenase (BCKDH), a rate-limiting enzyme in BCAA metabolism, potentially mediated by increased AMPD3 expression. Immunoblotting, in conjunction with proteomic analysis, revealed the presence of BCKDH not only in mitochondria, but also in the endoplasmic reticulum (ER), where it interacts with AMPD3.
