The brain's glymphatic system, a widespread waste disposal network, circulates cerebrospinal fluid to remove metabolic waste, thereby maintaining a healthy brain environment. MRI, along with ex vivo fluorescence microscopy of brain slices and macroscopic cortical imaging, currently represent the prevailing approaches for assessing glymphatic function. Despite these methods' contribution to our understanding of the glymphatic system, new techniques are needed to effectively address the specific shortcomings of each method. We utilize SPECT/CT imaging, coupled with [111In]-DTPA and [99mTc]-NanoScan radiotracers, to evaluate glymphatic function in different anesthesia-induced brain states. Utilizing SPECT, we corroborated the existence of brain-state-specific disparities in glymphatic flow and elucidated how brain states influence CSF flow kinetics and CSF outflow to lymph nodes. A comparative analysis of SPECT and MRI in imaging glymphatic flow revealed similar patterns of cerebrospinal fluid movement in both techniques, though SPECT demonstrated a greater degree of specificity across a wider range of tracer concentrations. We conclude that SPECT imaging holds potential as a tool to image the glymphatic system, with its high sensitivity and diverse range of tracers making it a viable alternative for glymphatic research.

Despite its widespread use globally, the ChAdOx1 nCoV-19 (AZD1222) vaccine's immunogenicity in dialysis patients has received scant attention in clinical trials. In Taiwan, we enrolled 123 patients receiving maintenance hemodialysis, a prospective study. Following receipt of two AZD1222 vaccine doses, infection-naive patients were monitored for seven months. The concentrations of anti-SARS-CoV-2 receptor-binding domain (RBD) antibodies were measured before and after each dose, and 5 months after the second dose, alongside neutralization capacity against the ancestral SARS-CoV-2, delta, and omicron variants, as primary outcomes. Time-dependent increases in anti-SARS-CoV-2 RBD antibody levels were substantial, with a maximum value of 4988 U/mL (median) observed one month post-second dose (interquartile range, 1625-1050 U/mL). A 47-fold decrease in antibody titer was noted at the 5-month mark. One month after the second immunization, 846 participants displayed neutralizing antibodies against the ancestral virus, 837 against the delta variant, and 16% against the omicron variant, according to a commercial surrogate neutralization assay. In the geometric mean of 50% pseudovirus neutralization titers, the ancestral virus, delta variant, and omicron variant displayed values of 6391, 2642, and 247, respectively. Antibody titers against RBD correlated strongly with the ability to neutralize the original and delta variants of the virus.
